Autoclave XYZ Qualification: A Comprehensive Guide

This document details a step-by-step qualification process for the XYZ autoclave, utilizing statistical analysis in StatGraphics Centurion.OBJETIVOS:

• No-Load Operation Qualification: Evaluate the autoclave's performance under empty conditions through three standard cycles. This assesses temperature, pressure, and heat distribution within the chamber.
• High-Load Operation Qualification: Assess the autoclave's effectiveness when loaded with materials, using three standard cycles. This focuses on heat transfer within the load and the biological indicator's response.

This qualification process validates the sterilization process by thoroughly evaluating both the operation and performance of the XYZ autoclave.

Responsables

• Validations Area: Responsible for developing the qualification protocol, ensuring results meet acceptance criteria, conducting follow-up actions, and delivering reports.
• Microbiology Area: Ensures the execution and adherence to protocol parameters related to the biological indicator.
• Production Area: Reviews compliance with protocol parameters.
• Quality Assurance Area: Approves the execution of qualification protocols.

Criterios de Aceptación

The autoclave must meet the following criteria to be considered qualified:

• Installation & Calibration: System must be installed according to design specifications, manufacturer recommendations, and BPMv guidelines. Instruments must be calibrated, verified, identified, and registered in the metrology and preventative maintenance program.
• Control System Functionality: All controls and alarms must operate as designed.
• Operational Range: The system must operate within specified design parameters or operational ranges.
• Vacuum Pump Performance: The vacuum pump must extract the chamber volume to the defined pressure and speed.
• Depressurization Rate: The average chamber depressurization rate must align with manufacturer recommendations.
• Cycle Parameter Security: The control system must prevent modification of sterilization cycle parameters through supervision or access codes.
• Sterilization Parameters: Temperature and pressure during the stable sterilization time must indicate 100% saturated vapor.
• Timer Functionality: The controller must reset the timer latency or cease temperature increase when a significant temperature drop occurs below the programmed level.
• Temperature & Pressure Uniformity (Operation Qualification): During the stable sterilization period, the temperature indicator must show a maximum average difference of ±1°C, and the pressure indicator must show a maximum average difference of ±1 PSI. The average temperature uniformity across all points during the stable sterilization period must not exceed ±1°C.
• Biological Indicator Results (Performance Qualification): Calculated F0 values for each evaluated point must be greater than 12 with a 95% confidence level. No growth of the biological indicator should be observed after the incubation period.
• Air Filter Integrity: Air filters must pass integrity testing.
